Chemical Property/Description

Bordeaux red to dark red or brown powder,Ponceau S is negatively charged and can bind to positively charged amino acid residues, while ponceau can also bind to non-polar regions of proteins to form red bands. The staining of proteins by Ponceau S is reversible and can be washed away with distilled water, PBS, or other appropriate solutions.

Main Uses:

Biological staining, protein staining after electrophoresis. Ponceau S can be used to detect proteins attached to NC and PVDF membranes, but cannot be used for Nylon membranes.

Determination of Cu in food by ponceau S probe dual wavelength absorption spectrometry. In TRIS-hydrochloric acid buffer medium at pH 6.86, ponceumin S reacts with Cu(2) to form an ion association with an obvious positive absorption peak and a clear negative absorption peak in the visible region. The positive and negative absorption wavelengths are 445 nm and 510 nm, respectively. Cu(2) follows Lambert-Bier's law in 0.06~1.6 mg/L(positive absorption, 445 nm), 0.04~1.6 mg/L(negative absorption, 510 nm) and 0.03~1.6 mg/L(DWO-VIS, 445 nm+510 nm). The apparent molar absorptivities (κ) were 8.70× 10-3 (positive absorption), 2.04× 10-4 (negative absorption) and 2.92× 10-4 (DWO-VIS)L/(mol·cm), respectively. The limits of quantitation were 0.62, 0.38 and 0.26 mg/100 g, respectively. The effects of absorption spectra, suitable reaction conditions and co-existing substances were also discussed. The dual-wavelength method was used to determine Cu in biscuits, noodles and milk powder. The recoveries were 97.9%-102% and the relative standard deviations (RSD) were 2.5%-2.8% (n=5).
